Tetra-amelia syndrome

Tetra-amelia syndrome is an ultra-rare congenital disorder in which all four limbs fail to form, usually accompanied by severe craniofacial, pulmonary, genital and pelvic malformations that make it lethal in utero or in the newborn period.

It is genetically heterogeneous, with biallelic loss-of-function variants in WNT3 or RSPO2 explaining the described families and a small number of other limb-reduction loci producing overlapping phenotypes.
