Temple syndrome

Temple syndrome is a rare imprinting disorder caused by loss of the paternal contribution of the 14q32.2 region, most often through maternal uniparental disomy of chromosome 14, and characterized by prenatal growth restriction, hypotonia, feeding difficulty, short stature, small hands and feet and early puberty.

Diagnosis rests on clinical recognition supported by methylation and copy-number testing of 14q32.2, and management is multidisciplinary, with growth hormone for short stature, gonadotropin-releasing hormone analogues for central precocious puberty and lifelong surveillance for obesity, dyslipidaemia and type 2 diabetes.
