Parkinson-plus syndrome

Parkinson-plus syndrome, also called atypical parkinsonism or disorders of multiple system degeneration, is a group of neurodegenerative diseases that combine classic parkinsonism with additional features that separate them from idiopathic Parkinson disease, namely multiple system atrophy, progressive supranuclear palsy, corticobasal degeneration and dementia with Lewy bodies.

The four disorders split into synucleinopathies, in which alpha-synuclein accumulates, and tauopathies, in which four-repeat tau accumulates, and they share a poor or short-lived response to levodopa and faster progression than Parkinson disease.

Pooled prevalence is about 3.6 per 100,000 for multiple system atrophy, 6.9 per 100,000 for progressive supranuclear palsy and 3.9 per 100,000 for corticobasal syndrome, against 100 to 300 per 100,000 for Parkinson disease, and no disease-modifying therapy is available for any of them.
