AOP-208

AOP208, formerly known as LB-208, is an investigational orally administered small-molecule antagonist of the serotonin 1B receptor (HTR1B) developed for oncology by the Austrian company AOP Health together with Leukos Biotech, a spin-off of the Josep Carreras Leukaemia Research Institute in Barcelona. It is described as a first-in-class agent that blocks a particular conformation of the 5-HT1B receptor on cancer stem cells, a target that had not previously been exploited in cancer therapy and that controls metabolic pathways involved in tumour cell survival and treatment resistance. A first-in-human phase 1 trial in relapsed or refractory solid tumours and lymphoma, SERONCO-1, is being run at the Vall d'Hebron Institute of Oncology in Barcelona, and a phase 1/2 trial in relapsed or refractory acute myeloid leukaemia and high-risk myelodysplastic syndrome, SERHEMA-1, began treating patients in 2026. The compound has no approved indication in any jurisdiction, and interim pharmacokinetic data reported in 2026 show dose-dependent exposure without plateau, supporting continued dose escalation.AOP208(原名 LB-208)是一种在研口服小分子 5-羟色胺 1B 受体(HTR1B)拮抗剂,由奥地利 AOP Health 公司与巴塞罗那 Josep Carreras 白血病研究所的衍生企业 Leukos Biotech 联合开发。
