ACBI3

ACBI3 is an investigational small-molecule PROTAC degrader that eliminates oncogenic KRAS protein rather than merely inhibiting it. Developed jointly by Boehringer Ingelheim and the University of Dundee, it recruits the VHL E3 ubiquitin ligase to KRAS and rapidly degrades 13 of the 17 most common KRAS mutants as well as wild-type KRAS, while sparing HRAS and NRAS. The compound was described in a 2024 Science paper and distributed freely to researchers through the Boehringer Ingelheim opnMe portal, but it has not yet been evaluated in patients. Its degradation-based mechanism produces more potent and more durable MAPK pathway suppression than non-degrading KRAS inhibitors in preclinical models. KRAS mutations occur in 17 to 25 percent of all cancers, so a pan-mutant degrader addresses the majority of KRAS-driven tumours that approved mutation-specific inhibitors cannot reach.ACBI3 是一种在研小分子 PROTAC 降解剂,它消除致癌 KRAS 蛋白而非仅仅抑制其功能。
